EC50 IC50 and Kd in Peptide Research

Three distinct measurement lenses surrounding one abstract molecular bead chain.

EC50, IC50, and Kd describe different properties of a measured system. Treating them as interchangeable can make a peptide comparison look precise while changing its meaning. The right interpretation starts with the endpoint, the experimental conditions, and the model used to estimate the value.

EC50, IC50, and Kd describe different properties of a measured system. Treating them as interchangeable can make a peptide comparison look precise while changing its meaning. The right interpretation starts with the endpoint, the experimental conditions, and the model used to estimate the value.

These parameters describe research measurements. They are not instructions for human or animal dosing, and they do not establish the performance of a separately supplied material outside the reported assay.

Understand what each parameter describes

EC50 usually describes the concentration associated with half of a defined maximal response in a concentration-response experiment. IC50 describes the concentration associated with half inhibition of a specified signal or process. The definition of the response range must be clear in each case.

Kd is an equilibrium dissociation constant describing binding under the assumptions of the model used. It is not simply another name for a functional half-maximal concentration.

The Assay Guidance Manual on data standardization explains distinctions among response parameters, including absolute and relative half-maximal values. Record the actual definition used in the source method rather than inferring it from an abbreviation.

Read the curve with the number

A fitted value is most informative when the underlying data cover a useful response range. Check the baseline, plateau, slope, and uncertainty. If the experiment does not resolve a plateau, the fitted half-maximal value may depend heavily on model constraints.

For a hypothetical example, Material A and Material B might share a reported EC50 while reaching different maximum responses. The matching numbers do not make the response curves equivalent. Conversely, two methods could report different EC50 values while using different definitions of the response range.

Retain full curves whenever possible. A single spreadsheet cell cannot show a poorly constrained fit or an excluded measurement.

Preserve units and logarithmic conventions

Compare values only after checking the units. A concentration in nanomolar units cannot be compared directly with a number in micromolar units. Keep the original value alongside any conversion.

Also check whether a result is reported as a concentration or a logarithmic parameter. For example, pEC50 is conventionally the negative base-ten logarithm of EC50 expressed in molar units. Under that convention, a larger pEC50 corresponds to a smaller EC50.

If a database stores both forms, document the conversion rule and avoid mixing the columns during sorting. A reversed ranking can result from a simple labeling error.

Match the experimental context

An IC50 depends on the assay conditions and should not automatically be treated as a binding constant. Binding parameter interpretation also depends on appropriate experimental conditions, as discussed in the receptor binding guidance.

For cross-study comparisons, record the target, species, cell system, readout, timing, and relevant method conditions. Where the experiments differ, present the values with those qualifications. A ratio assembled from unlike methods may not support the mechanistic conclusion suggested by its precision.

When the comparison matters to a project decision, a matched experiment may be more informative than adding more indirect literature values.

Build a useful parameter record

A practical record includes the parameter name, definition, value, unit, uncertainty, method, and source. Add the number of independent experiments and any constraints applied during fitting. State when a value could not be reliably estimated.

For laboratory purchasing, connect the planned assay to the specific material record rather than treating a published potency number as a product guarantee. Review the quality information and ask about available documentation. The laboratory should establish the evidence needed to interpret its own experimental parameters.
